質問者が納得そのアンプがヘッドフォン用の「サラウンドプロセッサー」を内蔵していれば可能です。
しかし、実際にはダメなものが多いです。
そもそも「5.1chサラウンドヘッドホン」は特殊なもの（東南アジアブランド：全部で6つのヘッドフォンユニットで構成）を除けば、それ自体は普通のステレオヘッドフォンです。
ソニーその他が発売しているサラウンドヘッドフォン・システムは「プロセッサー＋ヘッドフォン」で成り立っています。

デフォルトで付いてくるヘッドホンがゴミレベルなので、代わりにビクターのＨＰ－ＲＸ９００をＭＤＲ－ＤＳ１０００に付けています。
やはり、疑似サラウンドということでＣＯＤをプレイしているときも前後の音の判別は付きにくいと思います。
ＣＯＤをいつもプレイしていますが、足音で何となく後ろかなと思って、振り向いて正解だったことは多々あります。

ゼロであるならば誤検知が多くて有名です。
心配であるならメーカーに問い合わせるのが一番だし

質問者が納得もしオーディオマネージャの「5.1ch or 6ch（製品により表記が違います）」のサウンドチェックを試して音が出ない場合、割り振りが間違っている可能性があります。
その場合、オーディオマネージャのコネクタにカーソルを当てると何が割り振られているか表示されるので確認してください。色と割り振りの一覧は以下の通り。
緑：フロント（前面）スピーカー
黒：リア(背面）スピーカー出力
オレンジ：センタースピーカー／サブウーファ（重低音）出力
ピンク：マイク入力
